This tripeptide, composed of cysteine, glutamic acid, and glycine, is present in most mammalian tissue. It acts as an antioxidant, free radical scavenger, and detoxifying agent.
It is also an important cofactor of the enzyme GSH-peroxidase, essential for amino acid metabolism and leukotriene synthesis. As a substrate for glutathione S-transferase, it reacts with various harmful species, such as halides, epoxides, and free radicals, forming inactive and harmless compounds. In erythrocytes, these reactions prevent oxidative damage by reducing methemoglobin and peroxides. It is also involved in the formation and maintenance of protein disulfide bonds and the transport of amino acids across cell membranes. Cellular GSH content decreases with aging, even in the absence of disease, resulting in an increased oxidative state that appears to be related to reduced synthetic capacity in the second stage of GSH anabolism (catalyzed by glutathione synthase), without alterations in the metabolism of γ-glutamyl trans-peptidase or the restorative capacity of glutathione reductase in its antioxidant form, although this mechanism has not yet been fully explored in humans.
