Nrf2 (NF-E2-Related Factor 2), a human transactivator encoded in the NFE2L2 gene, regulates the expression of a set of detoxifying and antioxidant genes that protect the body from the ravages of oxidative stress. In a non-stress state, Nrf2 is anchored in the cytoplasm by its specific inhibitor, Keap1 (Kelch-like ECH-Associated Protein 1).
Keap1 functions as a sensor for oxidants and electrophilic xenobiotics. In the presence of either of these substances, Keap1 abandons its inhibitory function and releases Nrf2, allowing it to accumulate in the nucleus and bind to the ARE (Antioxidant Response Element) located in the enhancers of target genes. Under these circumstances, Nrf2 promotes the release of a variety of antioxidant enzymes and detoxifying proteins.
The aforementioned nuclear factor erythroid-derived factor 2-related factor 2 (Nrf2) (the primary cellular defense against the cytotoxic effects of oxidative stress), a member of the Cap "n" (CNC) basic region-leucine protein (bZIP) family, thus plays an important role in mediating ARE (Antioxidant Response Element) gene expression. Furthermore, research has shown that Nrf2 is an important modulator of susceptibility to carcinogen-induced carcinogenesis. The cytoprotective role of Nrf2 has been demonstrated in the susceptibility to cancer-eliminating mice; therefore, activation of Nrf2 signaling and induction of its target genes make it a nutrigenomic target of choice for cancer prevention. Furthermore, many phytochemicals are strong activators of Nrf2, regulating cellular defense enzymes through activation of the Nrf2 signaling pathway.
Sulforaphane (SGS), a naturally occurring isothiocyanate derived from cruciferous vegetables, induces cytoprotective phase 2 enzymes, supporting the body's response to oxidative stress and inflammation. SGS may modify critical cysteine residues of Keap1, leading to the stabilization and activation of AREs through Nrf2, inducing phase 2 enzymes. Research has shown that sulforaphane, through Nrf2 induction, protects the brain against hypoxic-ischemic injury and may improve cognitive function when administered after traumatic brain injury.
